Project Description: Municipal sewage sludge processing, utilization and disposal are of the most difficult and expensive operations conducted by municipalities today. In addition to the traditional agricultural application of biosolids and composting, recent trend in value-addition has seen another dimension where it is utilized for production of various biotechnological products, namely, biopesticides, bioherbicides, enzymes, bioplastics, biofloculants and biofuel. Value-added product implies that the value of the final product should exceed the cost of processing. The emerging value-added products are at different levels of production facing challenges in terms of process conditions, yield, scale-up or formulation. In this concern, we have developed process for the commercial production (biopesticides, bioplastics, biodiesel & biocontrol agents) using sludge as raw material, as described below. The production process involves following steps: sludge pretreatment, inoculation with required microbial strain, fermentation, product recovery, formulation, and field application.

Biopesticides: Bacillus thuringiensis (Bt) is the most successful biopesticide as of today being used actively in agriculture, forestry and public health sector. Bt is produced using expensive soya based synthetic raw materials. We have developed a low cost Bt production process using activated sludge as raw material. The sludge based biopesticides meets all the requirements of the biopesticides similar to a conventional process. Simultaneously, Bt also produces alkaline proteases which are in the soluble phase of the Bt fermented sludge.

Biodegradable plastics: Polyhydroxyalkanoate (PHA) is one of the biodegradable plastics produced mainly by bacteria and it has similar properties to conventional plastics.

A novel process of production of bioplastics (Poly(3-hydroxyalkanoate) or PHA) was developed employing activated sludge (AS) from full scale municipal, pulp & paper, starch, and dairy wastewater treatment plant, respectively. Low cost of bioplastics production (1$/kg versus 6$/kg in a conventional biological process).

Trichoderma: Trichoderma based biocontrol agents (BCAs) have added advantage of beneficial plant growth promotion activity compared to their counterparts (virus, bacteria, nematodes, and protozoa). Trichoderma has also been used for soil remediation of organochlorine compounds.

We used waste activated sludge to produce spores of Trichoderma. The sludge produced spores of Trichoderma viride were tested against plant pathogen (Fusarium sp.- a plant pathogen) for different crops (tomato, pepper and soya) and were found to be very effective. The mass balance calculations showed that to produce T. Viride required treating 1 hectare of field crops will require in order 100 kg of wheat or rice in solid state culture whereas same quantity of T. viride can be produced using 50 L of waste activated sludge with solids concentration about 30 g/L.

Bioflocculants: Sterilization, alkaline-thermal and acid-thermal treatments were applied to different sludge solids concentrations and the pre-treated sludge was used as raw material for Serratia sp.1 to produce extracellular polymeric substances (EPS). Maximum flocculation activity of 79.1% and increased dewatering by 52.2% was achieved using broth and crude capsular EPS, respectively. The results demonstrated that EPS having high flocculating capability could be produced using wastewater sludge as sole raw material. 

Biodiesel: We evaluated the energy balance and GHG emissions of biodiesel production from microbial and wastewater sludge oil. The results show that when oil extracted from municipal sludge is used for biodiesel production, the energy gain is up to 29.7 GJ per tonne biodiesel produced. It also shows that biodiesel production from sludge oil is a net carbon dioxide capture process.

Conclusion: The cost of production is 40 to 60% lower than conventional process and the product quality is similar to the industry standard. This research also provides a new alternative and sustainable method for sludge disposal. In the end, biotechnological processes will be commercialized with the help of participating commercial partners. The project’s benefits are threefold: i) a significant contribution to sustainable waste management; ii) the production of alternative value added products; iii) a reduction in greenhouse gas emissions by recycling the carbon contained in wastes.

The Microbial strains are ubiquitous in nature. Particularly, the sludge isolated strains are very potential and possess better ability to produce value added products over other microbial strains isolated from other environments. We have isolated many groups of microbial strains from sludge to produce value added products using sludge as raw material. These strains are found to enhance the bioconversion of sludge. Eleven PHA and PHB (bioplastics) producing bacterial strains were isolated from both municipal and pulp-paper wastewater sludge. These strains were found to accumulate the PHB granules. These strains were preserved and the gene sequences were submitted to Genbank, USA.

More than twenty-five extracellular polymeric substances (EPS) producing microbial strains were isolated from municipal sludge and found them performed well in sludge settling, when the individual strains are used bioflocculation of sludge. Further, these EPS producing microbial strains were also found to accumulate the bioplastics (PHA & PHB) during their growth in sludge.
